I am going to put 1x6 knotty cypress on the walls and ceiling of living/dining rooms. I am wanting to know if when I install it on the ceiling if 2 ft. apart nailing is enough. since that is what the trusses are centered on, the wood is tounge and groove also.If not what are my options? Also on the walls does it need to butt on studs? Could I cut the ends for splines to save waste if it does?

Having said all that. Four walls and a ceiling out of knotty cypress may be a bit much for 1 room.

I think it sounds a bit much too. But.. you might try running a bead of contractors adhesive down the middle of every 4th to 5th board. It will help them hold. I built a shed and used TnG 1x redwood for the siding. I put 3/4 plywood behind it so I could put nails where ever I wanted. That would be overkill but you might think about 1/4 inch.
