Door size VS opening .....
I tend to work closer than typical, when making a rough opening, allowing 1/4" on either side, sometimes a bit less. I don't use casings on the Craftsman style doors and openings because they don't look right to me. I just drywall right up to the jams as close as possible then run a bead of caulk.
So, if you glue up the jambs, there will be some flexibility, but not much. Obviously, the door will be square and the jams can be
pre-assembled or made in place.... I find it easier to pre-assemble them. If you chose to attach them in place, they should be plumb and allow for the door size in width. Depends on the style and whether there will be casings to cover the gaps.
The answer to your question will only be as detailed and specific as the question is detailed and specific. Good questions also include a sketch or a photo that illustrates your issue. (:< D)
